Age Related Changes in Small Intestinal Enzymes and Hepatic Antioxidant Enzymes in F344 Rats

Abstract
The current study was carried out to elucidate age-and site-related differences of intestinal epithelial enzymes and age associated changes in hectic antioxidant enzymes. Five male F344 rats per group were sacrificed at the age of 3wk (pre-weanling), 5wk, 10wk, 20wk, and 40wk to examine specific activities of akaline phosphatase (ALP), ¥ã-glutamyl transpeptidase (¥ã-GT), sucrase, maltase, and lactase in the small intestine and glutathionine-S-peroxidase (GSH-PX), glutathionine-S-transferase (GST), and superoxide dismutase (SOD) in the liver. The most significant increases in weight and length of the small intestine were achieved by rats aged between 3wk and 5wk. The mucosal tissues from 5wk old rats had significantly higher specific activities of ALP and ¥ã-GT than did tissues from the other ages. Regarding rite differences in ALP and ¥ã-GT, regional gradient along the length of the small intestine was remarkable (p$lt;.001; duodenum$gt;jejunum$gt;ileum). It was apparent that the epithelial tissues from pre-weanling rat (3wk old) exhibited the highest activity of lactase among all age groups. Conversely, for sucrase and maltase the 3wk old rats showed lower activities of these enzymes than any post-weanling rats. There was a significant effect on site in post-weanling rats, with activities higher in the jejunum than duodenum and ileum for lactase. The activities of hepatic cytosolic SOD and GSH-PX were not affected by age, whereas the activity of GST in rats aged 40wk were enhanced compared with those of rats aged 3wk, 5wk, 10wk and 20wk. It is concluded from the results presented here that development of intestinal eznymes is influenced by age and site and hepatic GST is also affected by age in F344 rays.
